Can use your mouse in right hand even when the comfort design says left?

You should be able to, its just not going to feel very comfortable if its designed for left hand

I have friends who do this because of various reasons including mobility limitations.

Windows allows you to swap the buttons if it makes your life easier.  If this is a new purchase, go to the store and put your fingers around the mouse to see how it feels.  Some micro will feel better than others.
