A bit more information would help us out in giving you an accurate answer. Are you asking how to set up an Xbox LIVE gamertag, or are you asking how to actually set up a connection to LIVE through your console?

Depending on what you mean,
If you don't have an account yet, you will need to buy a gold membership if you want all the benefits.
A one year membership usually costs around $59.99.
There should be a information on the back on the back with a PIN on how to setup.
Now if you already have our account set up, there is one of two ways to get your connection set up.
You can buy the wireless adapter which usually runs about $69.99-$99.99 The latest one is the Wireless "N" which has two antenna's. It's the fastest but you can probably get a better deal on a "G".
You will have to hook it up on the back of your 360, it goes into the USB port and clips on the back. Once its hooked up you should go into network settings and go from there.
The other way is through an ethernet cable which hooks up to your modem and plugs in the ethernet port on the back of your 360. Then again go into network settings under system settings. If it doesn't work it should tell you to restart your modem and turn it back on then wait 30 seconds and try again.

Note that the Wireless N adapter for the 360 will only be able to realize its maximum potential if you are also using a Wireless N router. If you aren't, you're better off getting the original one. The N adapter will still work with a G router, but you will only get the speed and range of the router you have.
Personally, I use a wired connection, because my Xbox is about five feet from my router. Depending on your set-up, that's all you'll need too. For the wired connection you don't need to change anything on your XBox's network settings. The console should automatically detect that there is a connection to the internet.
I'm not sure about the set-up for the wireless adapter, but I can't imagine it's much different from setting up a wireless connection on a PC. Go to network settings and choose your network, then enter the security code or password or whatever you've got for security key on your router (hopefully you've got something!) and then you should automatically connect anytime you turn on your 360 and sign in after that point - providing, of course, the router is actually connected.

Using the menu to the left Under connecting to Xbox Live you can select the option you wish to use. If your not sure what type of connection is best for you just select getting started from that menu and you will see a table with the pros and cons of each.
